LouieB
Oh hell yes. Goin' Down The Road Feelin' Bad - Good Ol Grateful DeadFor sure...and if you work at it you can find many versions of this song and just play this one song over and over by diffeerent artists... I once did a mix tape of all the songs that refer to roads and highways. That was kind of fun. Lots of blues and country songs talk about and name different roads and highways. His Mystic Valley Band stuff has a great modern americana vibe to it Josh Ritter: Various ritter tunes are great for road trippin. Most notably, Harrisburg, and Me & Jiggs. His live album is excellent Any live Grateful Dead Ryan Adams: Heartbreaker
Gobias Industries
personally i would just want to listen to Bob Dylan the whole time. This is generally a correct answer.
Golden Smoghead
I really enjoy Ben Harper and Steve Earle while on road trips. Especially early Earle, when he's still writing troubadour songs.

Guest Francis X. Hummel
I'll second the Dylan and Son Volt nods. I'd grab the entire Uncle Tupelo catalog as well. Listen start to finish.
Synthesizer Patel
you should end the mixtape with this: i've been everywhere
m_to_the_c
If you do make a city themed mix, by law, you have to start with "Washington, DC" by the Magnetic Fields. But I'd go for a mix of albums, not a mixtape. Depends how well you know her, I guess.
